Effectiveness of fluorescent pseudomonads strains HF22
and HF142 to control bacterial canker of tomato

Abstract
Two fluorescent pseudomonads strains, HF22
and HF142, were isolated from the rhizoplane of tomato roots, originating
from Souss-Massa-Darâa Valley, Agadir, Marocco. The potential utilization
of these strains to protect plants of tomato (cv. Daniela, Spain) against
bacterial canker was investigated. Tests were realized with spontaneous
resistant mutants to antibiotics: ampicillin (100 µg ml-1),
nalidixic acid (100 µg ml-1) and
chloramphenicol (25 µg ml-1).
Several treatments were tested to adhere cells of bacteria to tomato seeds:
distilled water, NaCl 0.85%, agar at 0.1% and 0.2%, xanthan gum at 0.25%
and 0.5%. The best bacterization was obtained with xanthan gum at 0.5%
as adhesive. The population fixed was ~ 4x105
cfu seed-1 and viable population after
28 days of storage of treated seeds at 5°C was ~105 cfu seed-1
for HF22r and ~5x10³ cfu seed-1
for HF142r. Tests of root-colonization
showed that HF22r and HF142r
established a large spermosphere populations (~ 3.8x105
cfu seed-1 for HF22r
and ~ 9x105 cfu seed-1
for HF142r), and colonization of tips
of roots was similar for both strains (~10³ cfu cm-1).
Control of bacterial canker under greenhouse in commercial-like conditions
using tomato (cv. Daniela, Spain) showed that the two strains significantly
reduced (P<0.001) infection compared to the untreated control.
The bacterium HF142 reduced the infection totally when applied as seed
treatment followed by a root treatment before transplanting compared to
the control.
